I'm making hydrogel particle for cell capsuling by collagen-chitosan
cross-linker is only NaOH.
not only I can't use any other cross-linkers, also can't add more NaOH in this task. (because there is any potential toxicity and I was not even allowed to test cell viability)
anyway I made Collagen-Chitosan 5-5mg/ml hydrogel particle.
but they aggregate together and I can't separate from oil because they are too weak strength after gelation (37℃). almost all particles are degraded.
when I was trying to mix for a thoroughly long enough time (1min ~1min 30sec by micro stir bar), it does not gel.
so is there any advice for improve strength of Hydrogel particle in oil
or
separation method particles from oil very mildly.
Try this: first disperse the powders into the oil with a rotor-stator at least 3000 rpm, add NaOH to the water phase in the correct quantity, and then add slowly the oil phase to the water phase, always stirring.
In this way you avoid agglomaration, as the gel hydrates.
