I aim to detect and identify nitrated and nitrosylated proteins in a particular cell type. These modifications, especially nitrosylation, is highly reversible. I have following 3 questions:
1. Is there a way to treat cells with "something" so that the proteins dont lose nitration/nitrosylation during time point collection, lysate preparation, and IP?
2. are there any good antibodies to IP nitrated Tyrosine, Cysteine, Tryptophan ?
3. Except Biotin switch, any other good method to detect nitrosylation?
