I have done 16 S rRNA sequencing of some of my bacterial isolates and one of my isolates is showing only 90% sequence similarity with reference strain as the maximum identity. Should I perform biochemical tests or is there some other way to identify the genus and species of this bacterial isolate?

It is long enough. How many times did you sequence your 16S rRNA gene for this strain? If it has been done only once, do you have the opportunity to confirm this sequence again? If yes, and assuming it is still giving 90%, then it might be a new species (since the % is not reaching the 97% threshold)

This value is very lower than the 98,65% (the novel threshold, Kim et al. (2014), then it is a novel species. In addition, it might represent a new genus or a new family. To determine the taxonomic status of your strain you should perform physiological and chimiotaxonomical study. The comparaison of 16S rRNA gene signature nucleotides in your strain and the closely related genera and families is also required. Please refer these articles. I hope this help you.
