I would have to go for familial studies in my research. I would include them in my thesis and research paper , and mean pedigree analysis as well. So please guide me how to conduct it? Have to get sampling from the siblings and parents and grand parents and the patients history about that specific disease? Please guide with all details.
There are two sides of conducting such a research:
1)Ethical side: You need to have proper ethical declaration/consent form approved by your ethics board. These studies would have to agree with the Declaration of´Helsinki. Also if the study involves some pharnaceutical/associated intervention than this requires identification of proper placebo group and mention of the risks etc. in the consent form.
2) Scientific side: You need to define inclusion/exclusion criteria. You need to define your controls(also if they need to be age/sex matched). The sample size for your study also needs to be determined and the power of the study defined using statistical measures. The time of sample collection/nature of sample/repeat measurements all need to be defined beforehand. Also you need to have a patient/control information sheet where you note down all basal clinical characteristics of the sample population which you are collecting.
It would be more helpful if you define your study in more exact sense then I can tell you what exactly you will need.
Family studies are a powerful way to identify loci linked to a particular genetic trait. If the goal of your study is to localize a trait to a particular chromosome, this is a great method. Start by very clearly delineating the phenotype you are working with. Once you have a phenotype, collect samples from as many individuals as you can, both affected and unaffected, and from as many generations as you can. Even if you don't have historical samples, it is desirable to develop a pedigree of the kindred to connect all patients to as few common ancestors as possible. Identify the trait status of each individual in the pedigree. Once you have your pedigree complete, you can begin the lab work - genotyping and analysis.
Tracey I am studying the area where we have patients who has a specific disease about 50 present of them have parents who has the same disease, so when i am studying these patients (not parents) is it the First Generation or the second as their parents were or are effected too. Or is it the Second Generation? And maybe I study the offspring of the patients so it would be third generation?
