I am trying to run Thin Layer Chromatography that targets terpenoid compound on an ethanolic extract of leaves. So I am using methanol : ethyl acetate as the eluent with the ratio 4 : 1. Actually, I've been trying to optimize the eluent, but in the end the best one is that I mentioned before. The problem is, the band is not well-separated. When it is run, terpenoid's just visualized as a line that look like a tail, without being separated (I sprayed it with Liebermann-Burchard reagent). Can somebody help me to figure out how to solve this problem? Thanks in advance!
The product may not be well soluble in this solvent, so you can try other solvents.
Sheva Rimma It is better to try the the different ratios of Hexane : Ethyl Acetate for the separation of Terpenoids. For example 6:4 or 7:3.
To improve TLC separation of terpenoids, optimize the solvent system, use alternative detection reagents, adjust chamber conditions, refine sample preparation, and select high-quality TLC plates.
