Hi I am growing marine algae however i have lots of contaminants in my samples such as rotifers . I have attempted to remove these with sodium bicarbonate however the eggs still do remain. i have tried filtration with a 0.45um filter however the filter cloggs. Any suggestions on how i can remove these without losing the algae as the aim is to have a pure culture?
A 0.45 um filter likely has a pore size that is too small to capture the rotifers yet let the algal cells pass through. Determine the size of the algal cells and select a filter with an appropriate pore size. Alternatively, you can do a dilution culture series to try to clean up your initial culture.
I've found dilutions and/or pipetting cells under an inverted microscope as the best ways to get a rough 'clean' culture.
