I will do sequencing of my sample. After checking my desired band by gel electrophoresis, I cut the specific band on the gel in order to do sequencing. I have followed gel purification kit and used "nuclease free water" at the last step for the elution(as elution buffer did not work well). I am not getting desired concentration and purity of DNA. The concentration always remains in between 2-10 ng/ul and 260/280 ratio is >2. I am so disappointed. I am also purifying my pcr product which purity is good. But I want solve the gel purification problem. Please suggest me to get good purity of DNA directly from the gel.
