I have started working on Hydrangea macrophylla. I need to do some experiments on seed priming and check results on seed germination. but hydrangea seeds are so small and also very difficult to handle. I have tried some basic methods but I failed. so please if anyone has expertise in germination of hydrangea seed. please share a protocol with me.
Use a petri dish with cotton or moistened filter paper and keep the seeds in a warm, dark environment, ensuring the substrate stays moist but not soggy, and regularly observing for signs of germination. But before that u need to disinfect with mercuric chloride.
- Seeds: Choose the type of seeds you want to germinate.
- Petri Dishes: Sterile petri dishes are ideal for controlled experiments.
- Filter Paper or Cotton: To provide a substrate for the seeds to germinate.
- Sterilized Distilled Water: To keep the substrate moist.
- Parafilm: To seal the petri dishes and maintain humidity.
- Incubator (Optional): To control temperature and humidity.
You need to determine first if the seeds are viable. You can use the flotation test. Seeds that sink in water are viable, those that float are not. then only use viable seeds. Then follow what Geetika Singh says :)

I have already did this method but it is not working because the seeds of H. macrophylla are so tiny . so going through literature I found that for such kind of seeds the best method is using a media containing petri dish (e.g. MS media). I hope that works for me :)))
