what techniques can be used to discover actual site of protein production? for example, human liver is the site of synthesis for hepcidin(protein). I would like to know how to conclude that the liver is site for synthesis?.
There are number of ways of getting the information. To some degree, it depends on the protein(s) of your interest and their abundance. Ideally, you would attempt to detect protein(s) in each organ/tissue that interests you.
You can probably begin with literature survey, including databases which already have information about expression of many proteins in a variety of tissues.

If your protein of interest is human protein, you may find "Human Protein Atlas" informative. Even if you protein of interest is not human protein, you may get some idea.
