This problem is called multi objective path planning. You can easily find many papers about.

Look for this paper: Multi-objective path planning in discrete space. Notice that in this paper they used grid, each cell could be a node in a grid graph (similar to your graph)!

In general, you may represent the chromosome based on the path and then search the space by applying genetic operations.

I prefer to use HeuristicLab software instead of Matlab. It is very efficient and includes many algorithms about industrial engineering topics. Beside, it is easy to use and has GNU licence.
