I want to extracting the RNA from MCF-7 cell that treated by an extract of plant, i a bit confuse to add the lysis buffer RNA, shoud i get the pellet cell first and adding the lysis buffer on the tube or should i adding the buffer directly on its culture plate? thank you before

If you pellet your cells anyway, why not resuspend your pellet in Trizol/Tripure for storage?
After resuspending in Trizol/Tripure your sample can be stored at -20 or lower for a very long time, since the Tri-reagent will neutralize the RNases present. This assures there will be no detrimental effects of freezing your cell pellets at -80.

just in case the first trial will not has a good RNA concentration
