Isolation of MRSA directly from milk sample may not be feasible. However, stepr-wise processing will help you to get MRSA from milk.
Firstly, the milk (diluted or neat) sample needs to be enriched with peptone water or BHI medium for overnight.
Plate the enriched sample on mannitol salt agar (selective medium for staphylococci).
Colonies that turns the media color from pink to yellow can be subjected for further biochemical and microscopic analysis.
Catalase, coagulase production is the typical characteristics of S. aureus.
The identified S. aureus can be subjected for antimicrobial susceptibility testing including oxacillin (for methicillin resistant strains).
Oxicillin resistant s. aureus strains can further confirmed by mecA/femB amplification.
