As to starch I recommend the validated enzymatic assay by K. Brunt and P. Sanders in Starch 50 (1998) 413-419. Unfortunately, I have only the Abstract available. For protein you can use the standard Kjeldahl method with conversion factor (%N to protein) = 5.7 generally used for cereals.
