The easiest way to get analysis of of infection by macrophomina before senescence and maturity is to stress the tissues with paraquat herbicide which over rides the natural resistance mechanisms and causes fungal expression based on nutrient release.
Dr. Ray Cerkausas and Dr. James B. Sinclair co authored this work which was done when I was studying in Sinclair's laboratory.
Paraquat is quite toxic and care needs to be taken to avoid contact. I would suggest using this paraquat technique along with selective media
O. Dhingra developed and then once the isolation was confirmed returning to a standard media.
Hi Pankaj : First think, you should have idea for the field that you use to plant with soybeans , if you have been had this problem before with soybean in the same field, you should expected to get the same problem with the next season.
The stress stage of the plant (flowering and pod filling) make the plant weak and easy to take with week fungus like Macrophomina spp, also too much water (irrigation) make this problem worst.
Macrophomina infection, but at the beginning of the growth of infected plants we notice elongation in the plant because of the pathogenic fungus indol acitic acide
