I want to do UV VIS of a silicon substrate,so is there any different sample holder required for that?can it be done on conventional UV VIS spectrometer where cuvette is used as sample holder
For silicon substrates, I advice you to do UV-Vis spectrophotometry using transmittance or reflectance mode (not absorbance). For the substrate holder, you cannot use the cuvette since it is not adapted to solid substrates. Instead, you can use an integration sphere where you can put your sample beteen a white transparent cube and the UV irradiation.
There is no absorption band of Si between 190 and 900 nm, either in transmission or reflection. If you want to know whether Si adds any background to a sample deposited on it, the answer is 'no'.
