I'm working with a crosslinked gelatin-based hydrogel system, where cytokines are embedded within solid gelatin slices. Heating the matrix to dissolve it could compromise the integrity of the cytokines, so I'm exploring alternative methods to disintegrate or extract cytokines from the gelatin without applying heat.
Are there any enzymatic, chemical, or mechanical approaches that can break down gelatin in solid form while preserving cytokine structure for downstream quantification (e.g., ELISA)?

To extract cytokines from gelatin without heat, you may use any of the non-thermal methods provided below.
1. a) You may incubate the gelatin in a suitable extraction buffer, which could be an enzyme solution containing gelatinase or pepsin; b) an acidified solution, acidic extraction is used for Type A gelatin, while alkaline extraction is used for Type B. Mild acid, like acetic acid, can be used at room temperature to swell and extract gelatin; c) certain detergents, like Triton X-100, are effective at disrupting protein-protein and lipid-protein interactions. This can help release cytokines without denaturing them, though detergents should be chosen carefully to minimize potential damage to the cytokines; d) you may apply high-pressure processing to break down the gelatin matrix, or use ultrasound-assisted extraction which uses pressure waves and cavitation to disrupt the structure to facilitate the disruption of gelatin and release of cytokines; e) repeated cycles of freezing and thawing can lyse cells and disrupt tissue through the formation of ice crystals. While commonly used for cell lysis, it can also assist in breaking down the gelatin matrix. This method is inexpensive but can be lengthy.
2. After completion of step1, centrifuge or use filtration to separate the cytokine-rich liquid from the remaining gelatin matrix.
3. You may further purify the extracted cytokines using immunoprecipitation or chromatography to isolate the specific cytokine of interest.

Malcolm Nobre, I am using gelatin to dissolve in RO water and EDC/NHS reaction to also dissolve separately in RO water to mix with gelatin for crosslinking. There, I would be lowering the pH with HCl to 6.0. Later, after hydrogel formation, I would be washing the hydrogel with PBS to stabilise the pH around 7.4, as my work has cells in the hydrogel. So, would this approach work fine?
Pratyush Sarin, yes, your approach sounds good. But you must verify that the acidic pH (6.0) does not damage the cells or affect EDC/NHS reactivity and confirm that the final PBS washing adequately removes unreacted EDC/NHS, which are cytotoxic.
