Dear all, 

Currently I am dealing with curcumin. I need to determine its MIC on C. albicans. I have searched a lot of the papers and I found that almost all of the researchers used the CLSI M27A as reference. According to the reference, RPMI 1640 medium is used as growth medium for fungi. 

As I know that, curcumin is very insoluble in water. I have tried to dilute 20 mg/ mL of curcumin stock (in 100% DMSO) to 1000 ug/ mL ~ 50 ug/ mL in RPMI 1640 medium (pH 7, buffered with MOPS), but I did saw there were precipitates formed at the bottom of the well for all of the concentration. 

As a result, does anyone know that what method did they actually used so that they could gain about 1000 ug/ mL of curcumin for determining the MIC of C. albicans?
