Currently I am dealing with curcumin. I need to determine its MIC on C. albicans. I have searched a lot of the papers and I found that almost all of the researchers used the CLSI M27A as reference. According to the reference, RPMI 1640 medium is used as growth medium for fungi.
As I know that, curcumin is very insoluble in water. I have tried to dilute 20 mg/ mL of curcumin stock (in 100% DMSO) to 1000 ug/ mL ~ 50 ug/ mL in RPMI 1640 medium (pH 7, buffered with MOPS), but I did saw there were precipitates formed at the bottom of the well for all of the concentration.
As a result, does anyone know that what method did they actually used so that they could gain about 1000 ug/ mL of curcumin for determining the MIC of C. albicans?

Thank you for your help. I have read the paper suggested by you. However, the medium used by the authors is not suitable for my experiment since I am dealing with C. albicans. The concentration used by the authors is lower than my desired concentration.

In some of my research, we used cyclodextrin as an alternative to DMSO. Since it's a small molecule, it can "hide" in the structure of cylcodextrin and become solubilized. However, I'll add as a note, while this has the potential to solubilize the compound, it might cause the MIC to shift, since some of the compound may be trapped in the cyclodextrin.
