I will load the cell free cholesterol, then I need extract the cholesterol from the cell. With WAKO kit to do lipid assay.
You should be able to use the Wako for the assay. Read-outs can be both by fluorescence or absorbance

First you should extract the lipids , B&D extraction
After the extraction you can resolve the lipids , triton tx2%, and use the kit for cholesterol determination

I agree with Vincent Bloks but the first question is :¿ Which kind of cell ? because the results are depending on the amount of cholesterol which is different f.e.in an adipose tissue cell or hepatocyte cell , human cell or murine cell in different tissues.
