I just started doing field stimulation in the CA3 region of wild-type (WT) SVE129 mice (2-4 month old) to record long-term potentiation in the CA1 region. However, I have major problems seeing LTP in young sve129 mice slices. Instead of an instant excitatory postsynaptic potential (EPSP) elevation, which is maintained at least 30min (typical LTP), I see a slow incline over 5 to 10 min. I already tried to improve my slicing, double checked my solutions and changed batteries in the stimulator. I would be grateful for your help and advice how to fix it.

I already use young adult mice (2-4 month old) and I need to stay in the adult system.
The initial response is the problem. It either drops or stays the same as baseline response for 2-4 minutes and then raises over `slowly` (within another 2-4 minutes) to 130-150%. So it is not typical LTP.
Sorry for the misunderstanding, but we record baselines for 20 min after an adaptation phase of 20 min.
I meant that right after the high frequent stimulation ... i should see 30-60% increase right away... but I do not, but see similar signal or less EPSP signal than when I record the baseline.
