Total chrome is determined by AAS. Cr(VI) is analyzed spectrophotometrically with diphenylcarbazide. Cr(III) is obtained from difference between total chrome and Cr(VI) concentration.
Muhammad... I think you are after following of the ability of some microorganism to convert the toxic Cr(VI) into the Cr(III). During the course of incubation you may draw samples at certain intervals, extract the mineral content with deionized water and take the extract to evaluate the progress of the conversion process by spectrochemical analysis. The total Cr content needs only to be determined in the start point of the process.... good luck.
