I focus on the conformational transition and function connection. The fluorescence energy resonance transfer system based on fluorescent probes is used to study conformational changes. The fluorescence lifetime of the probe limits the long-term dynamic detection in living cell. so, is there other methods suggested for me?

I think you may be referring not to the fluorescence lifetime (usually measured in nanoseconds) but to the stability of the fluorescence over longer periods of time due to chemical instability or photochemical bleaching.
Photochemical bleaching can be reduced by either reducing the exposure of the fluorophore to light or choosing a fluorophore that is less susceptible to photobleaching.
