you can use several tools to detect nonsynonymous SNPs on the exonic regions. such as SIFT, PATHER, POLYPHEN1 AND 2, PROVEAN, PREDICT SNP, SNP&GO, I-MUTANT2, FTSITE SEARCH, CONSURF.............etc.
