"For isolation of spinal motor neurons, lumbar parts of the spinal cords of E13.5 wild-type mouse embryos were harvested and meninges should carefully removed. Afterward, the spinal cord tissue was enzymatically digested in 0.25% trypsin/EDTA solution for 30 min at 37 °C. After gentle mechanical disruption of the tissue, spinal cord cells were pre-plated on an uncoated plastic culture dish for 1 h at 37 °C to remove most of the non-neuronal cells as glial cells or fibroblasts. After that, the cell solution should be removed from the dish and centrifuged at 500× g for 5 min. Next, the pellet was resuspended in DMEM: F12 1:1 containing 10% FBS, 1% P/S, and Glutamax. Next, cells were plated on PDL-coated glass coverslips (5000 cells per coverslip) and cultured at 37 °C and 5% CO2. At 1 DIV, the medium changed to Neurobasal A medium containing B27, Glutamax, P/S, GDNF, BDNF, CNTF, and 1 µM AraC. Afterward, the medium was then changed once a week. Cells keeep in culture until used in experiments."
