In in-vivo animal model, we calculate the dose in mg/kg body wt. How can we extrapolate the same in in-vitro evaluation? Is there any method or calculation to execute the same??
This is tricky. The best way to do it would be to examine the blood concentration of the drug over a period of time (which should help you determine a half life as well). If it's a commercial drug, this information should be available with the product. Once this has been determined, you can perform a dose response assay on your cells in vitro. Using gene expression (for example), you can measure a gene that should be responsive to your drug, and compare that to results you've obtained in vivo. Probably not the answer you want (it's a lot of work!), but I hope it helps.
