I have 37% formaldehyde solution (formalin).I m working in avian influenza vaccine production.
Many articles mentioned 0.01% ,0.02% formalin inactivate virus .
But
How does I converted into 0.02% formalin from 37% formalin.
Then howmany ml of converted formalin should I add into allantoic fluid?
in order to prepare 100 ml of 0.02% formalin you can use the following formula:
%C = volumen ethanol/volume of solution
(0.02/100)=((37/100)x/100) ===> 0.0002=0.37x /100
2=37x ====>x=37 / 2 ====> x=18.5 cc
it means you should add 18.5 cc of 37% formalin to 81.5 cc of distilled water; the final solution will be 0.02%
about the cc of this solution you should to allanotic, I have not definite idea and you should obey the articles.

To make 100 ml of 0.02% formalin, take 54 microlitre from your 37% formalin solution and make up the volume to 100 ml with your desired solvent medium(distilled water or others).

There has often been confusion between formalin and formaldehyde. Formalin is a saturated solution of 37% formaldehyde. When scientists publish articles they sometimes put formalin concentration, when it was formaldehyde concentration they had used. You have to find out what concentration of formalin or formaldehyde needed to inactivate influenza virus for preparation of vaccines. I think 0.01 % or 0.02% formalin (which will be 0.0037% or 0.0074% formaldehyde) is low for inactivation of viruses for preparation of vaccines. The logical and scientific way to verify the concentration of any inactivating agents, including formalin, for preparation of vaccines is to evaluate an inactivation curve of the virus by taking samples of your virus pool with inactivating agents, periodically (for example daily) and test for residual live virus. When you don't detect any live virus, you should add extra time for inactivation to ensure complete inactivation. These conditions should be used for inactivation of your vaccine. I think you should be able to find inactivation curves of viruses with formalin in published literature.
I think you got answer from above discussion. But important is the contact time of formalin with allantoic fluid. It should be 2 hours at 37 degree. Otherwise virus could show pathogenicity.
I think, 0.02% formalin dosage means you have to add 0.02 ml of formalin per 100 ml of Allantoic fluid.
