Dear Researchers,
A concentration of QE varies from 25, 75,125,175 to 225 µg/ml have been used to get the equation y=0.0008+0.0314, R2=0.999 from the standard calibration curve. The concentration of the plant extract in the assay was 25µg/ml. However, the total flavonoid contents seem very high (6402mg/gm) as it have been shown below.
The total flavonoid content was calculated using the formula: TFC=C*V/M,
Where, TFC: total flavonoid content,
C: concentration established from the calibration curve in mg/ml
V=volume of the extract used in the assay in ml,
Volume of extract used in the assay in ml(V)=1ml
Weight of dry extract per ml in gram (M)=0.000025g
Absorbance=0.15944
QE concentration in mg/ml (C)=0.16005mg/ml
TFC in mg/gm=6402 mg/gm
What is the problem with this high value? Hope to hear from you all soon.

how did you calculate QE? I am not able to reproduce your result.
(Mine is: ~200 mg/g)
Also your fitting equation is not complete, x is missing, maybe that is why my result is different from yours.
Your formula looks fine though.

It would be good to check the final concentration of the obtained extract starting from the weight of the raw material and the dilution as well as the QE concentration.

I missed the x in the fitted equation. Y=0.0008x+0.0314.
X was calculated as:
X=0.15944-0.0314/0.0008
X=160.05ug/ml or 0.16005mg/ml
C=0.16005mg/ml
Mass=0.000025gm
V=1ml
TFC=C*V/M
Tfc=0.16005mg/ml*1ml/0.000025g
Tfc=6402mg/gm
That is how I calculated the TFC.

Thank you.Here is how I calculate the extract in the assay.
Initially I kept the extract @ 50mg/ml
Then I took 0.15ml from the 50mg/ml stock and diluted into 5ml methanol. Then I got 5ml of 1.5mg/ml concentration. Then I took 0.4ml from 1.5mg/ml concentration I got a final concentration 0.6mg/ml concentration, which was used in the assay.
