We usually use spectrophotometric method to determine the loaded drug on a support. You have to check the concentration of the drug in the solution before loading and after loading to see how much of it is lost.The difference is the amount of loaded drug.
Unfortunately, it is practically impossible to calculate it theoretically as it depends on nature of a drug, nanoparticle and its components including shell, solvent, temperature and inevitably, on physico-chemical interactions (including parameters of solubility) of all components. But as Mohammad Kooti said it can be done through experimental techniques. I am sorry to disappoint you:)
