Having the results of the S21 (transmission coefficient) how one can deduce the received power? Or
What is the relationship between both parameters?

If the transmission co-efficient is defined as
S21= 2* (VLoad / Vsource)* (Rsource/ RLoad)^0.5

Where the parameters refers to the load voltage, source or input voltage, source resistance and load resistance respectively.
Then by rearranging the terms,
Output power, Pout = (Vout^2) / RLoad = { ( S21 ^ 2) * (Vsource ^2 ) } / (2* Rsource)
In my opinion, it would be a good option to cross check the mathematical analysis with simulation result to verify the accuracy of analysis.

Can you please tell me, in the case where I only have the gain in dBi of the Tx and Rx antenna and knowing that the S21 that I have is in dB, how can I use this formula?

Khadidja Moudjari you can use this formula to get the magnitude from the dBi value.
ydb = 20 log10( y ).
If you use CST software, there is an option to see the linear value under 1D plot.
