Hi, I need to count DAB positive cells in a tissue and eventually calculate percentage DAB positive cells in a given area like for eg. a gland. I tried using Fiji and Qupath. Qupath didnt seem to work well with me. Fiji was giving me different counts with different shapes of glands. Shape and size of glands at the same magnification may vary. However, the problem with Fiji is that parameters set for one gland does not give appropriate results with some other gland. And I have to keep the settings uniform throughout the analysis for uniformity. Either I am not doing it the right way or I need some other software for this kind of analysis. Please help me out with.

Thank you for the reply. I tried using QuPath software. However, despite following the tutorial regarding estimation of DAB positive cells in the tissue using QuPath, I was not able to get the positive count. Is there any link which i could use to go step-wise for the count?
