I have synthesized Copper nanoparticles using plant extract as reducing agent. I want to standardize the protocol for this green synthesis of CuNPs. For this I need to know approximate size of CuNPs so obtained using UV-Vis Spectrophotometer, before going for SEM and TEM. Please suggest how can I do so?
Also suggest what should be taken as reference or blank....? CuSO4 solution or plant extract or distill water or the reaction mixture at time 0 minutes

TEM is the definitely the best way to size your Cu NPs, allowing also the shape of the Cu NPs to be determined.
That said, should be able to calculate Cu nanoparticle sizes from UV-Vis spectra, based on the LSPR position and the absorbance at the localised surface plasmon resonance (LSPR) maximum. Both will be sensitive to Cu NP size, shape and dispersion medium. I suggest synthesizing Cu NPs in a range of different sizes, collect absorbance spectra, and then use that data to construct your own calibration curves of LSPR position versus Cu NP diameter and absorbance at LSPR maximum versus Cu NP diameter.
