I have done grinding of 100 mg leaf in 3 ml of 0.1% TCA. after centrifugation 1 ml of supernatant + 4 ml of 20% TCA containing 0.5%TBA was waterbathed at 95 0C for 30 min. then O.D was taken at 532 and 600 nm. from this how could i calculate MDA in mM/g FW ?
how tissue amount and total volume involved in its calculation?

Vous devez calculer la DO spécifique : DOsp = DO532 – DO600
En exploitant la loi de Beer-Lambert (Abs = ƐCL ; Ɛ = 155 mM-1 cm-1 ; L = 1 cm) vous pouvez alors déterminer la concentration du MDA : C (mM) = DOsp/155 ; ramener cette concentration en μM au besoin.
Comme votre mélange réactionnel est de 5 ml, vous multipliez la concentration obtenu par 5. Ne te reste plus qu’a ramener cette concentration au matériel de départ.
