Basically, I am analysing Fluorobenzoic acid using GC-NICI-MS single quadrupole system. The steps of quantification includes SPE + Derivatization but there are irregularities in the results hence no reproducibility are observed.
The derivatizing agent is Pentafluorobenzyl bromide and extracting solvent is acetonitrile. If anyone can suggest, how to perform linearity and extraction efficiency calculation.
Should I go for internal standard quantification or external standard and how to calculate the enrichment factor in the case of derivatization.
The best option is to work with an internal standard. This compund must be added before starting the extraction and derivatization.

It will be better if you please indicate whether an internal standard should be added in fix amount both in post spiked sample and before extraction sample.
What to do if the Extraction efficiency of internal standard is not 100%.Every time it will give the different response in GC-MS.
Should I choose the deuterated analogues as internal standard or already esterified compound? Please help.
