For large scale production, I had set the conditions similar to shake flask, but the cell growth had been restricted (evident from abs-600nm- 1.2). What could be the possible reasons?
Are you using plastic flasks that have been 'cleaned' with virkon? If not rinsed thoroughly several times, residual virkon can negatively affect future bacterial cultures, i.e. foaming and low OD growth. For this reason, we never use virkon in our plastic flasks unless there's an absolute need to use a harsh disinfectant (e.g. highly pathogenic bacteria culture). Just a thought, best of luck.
What are your conditions? you have to consider several factors to optimize large scale production. For example, shaking sometimes is not as effective in large containers as it builds up foam. Foam build up could be reduced by aeration in some cases unless you are growing anaerobs. If you are growing cultures that produce a lot of mucilage, then aeration is not your best solution.
