I prepared gelatin nanoparticles using two-step desolvation method. I used centrifugation to isolate nanoparticles but the problem is that the nanoparticles get aggregated when I redispersed them. I decreased the speed of centrifugation to 10000 g for 15 min, it worked perfectly but the pellet was very small almost nothing especially after the third centrifugation.
How can I avoid the aggregation without losing the yield of nanoparticles?
*I used type A gelatin bloom 300 to prepare nanoparticles.
*Speed of centrifugation (14000-16000 g) for 15 min/3x
Please try biospin comumns, it may help you to avoid aggregation.
Maybe the problem is that particles are highly hydrated, i.e. density difference is very low and reducing acceleration below critical level you will only able to separate the larger ones. You may think in direction of surface functionalization, e.g. by biopolymer complexes. You may look into work of group of McClements, which used this approach for emulsions.
