Hi, all. We are trying to evaluate the resistance to amphotericin B using budding yeast as a model system. I am wondering if anyone know that how long does it take for ampB to kill a wt yeast cell? We are using Propidium iodide to stain the cells. And currently we are using 3 hour AmpB treatment. I am wondering if anyone have a better idea on these two questions: (1) how long does it take for ampB to kill a fungal/yeast cell (2)if we use PI to stain the cells killed by ampB, how long should the treatment time be? Thanks.
