Hi, I have a question, I inoculated the fresh bacterial culture and incubated overnight in 5ml nutrient broth, I must centrifuge 4430*G for 5 minutes followed by decantating the supernatent and washing with PBS and centrifuging again. These steps are three times repeated and all the resuspended pellets were adjusted to a final volume of 10ml to achieve 10^9 CFU/ml. However my question is should I centrifuge the whole 5 ml of the overnight culture or can I just use 1 ml of it ? which is the best way? Can you please help me with this? My final goal is 10^9.
