I have powder from both Sigma and Himedia. Molecular weight 251.17. Generally, once the solution is prepared we use 5%-10% of the reaction volume such as 20µL Alamar Blue solution for 200µL reaction volume. But how to prepare the solution from commercial powder?

First you need to make a stock solution of 1% that is weigh 0.1 gram powder and add in 10 ml sterile water. Use final volume of 0.02% in your viability assay.
George, I have a question....since you use 0.0335g to 45mL DIH20 or PBS, that would give you a 0.074% solution. How is differ from the recommended of 0.02% in the testing wells? Is this too much?
Commonly 0.015-0.02% can be used; if 0.02% (0.02 gm in 99.8 ml of water), so that 0.02 = 99.8 ml
x = 10 ml >>> 0.02 x 10 ml/ 99.8ml= 0.002 gm= 20 mg
