Hello, I am going to transfect A549 cells by creating a mutant form of a protein.then I will create a spheroid model of these cells.for the spheroid model I am thinking of coating 24 well plates with 1% agarose.how many microliters of agarose should I coat with each well and how many thousand cells should I seed?
100-200 µl of agarose is needed in a 24 well plate. However, I warn you that the spheroids will be chaotic in size and shape if you just sow the cells on the agarose layer. It may be more convenient to make microwells in the gel, for example with 3d printed stamps.

I agree with Artem Minin on this. I have tried it once and the spheroid only looks good for 1-2 days. I think it would be better for you to purchase some ultralow attachment plate or microwells for spheroids in this case. But if you’re on a tight budget and time, you can try, just be sure to also autoclave the agarose instead of just microwaving it, to ensure no contamination is carried over. Good luck
