About how to test, CCK-8 Kit is convenient to test cell proliferation or viability.
thanks kelong ,could you tell me in detail about this test and how long period it takes to make resistant cell lines some papers have stated from 3 to 6 to 9 to 12 months,and can we freeze thaw cells once they reach IC50 conc.or the resistant cells should remain in culture till your experiments are done which is not feasible.
Oh, I am sorry. I didn't test a cell line for so long time. You may get the information from customer service of the company for CCK-8. Hope this would help you.
For the period the resistant cell lines takes, it depends on the cytotoxicity of drugs and the conditions of cells. If you get the resistant cell lines, you can freeze thaw them. But I do not know well about tamoxifen, and I sugget you buy or ask the authors of published papers for such cell lines. If you do not konw well about the procedure, it will take more time to get the cell line.
thanks i have been growing cell line from last 8 months,an d got MTT assay as well with IC 50 15uM only thing was incubator went bad ,so the running culture couldn't be saved ,i ha dto freeze thaw three months back culture in between again,now i am growing the cell and i have to check are they resistant.? for me as well it took 8 months to get the cells,hope they ares same now.that was my worry. rest i am in contact an d literature work by same area.thankyou .
I have the same problem too. But I using detection of P-glycoprotein expression to confirm the resistance of cell by using flow cytometer. Unfortunately the expression would not show even after 3 mounts of Tam treated cell
