For the period the resistant cell lines takes, it depends on the cytotoxicity of drugs and the conditions of cells. If you get the resistant cell lines, you can freeze thaw them. But I do not know well about tamoxifen, and I sugget you buy or ask the authors of published papers for such cell lines. If you do not konw well about the procedure, it will take more time to get the cell line.
thanks i have been growing cell line from last 8 months,an d got MTT assay as well with IC 50 15uM only thing was incubator went bad ,so the running culture couldn't be saved ,i ha dto freeze thaw three months back culture in between again,now i am growing the cell and i have to check are they resistant.? for me as well it took 8 months to get the cells,hope they ares same now.that was my worry. rest i am in contact an d literature work by same area.thankyou .
I have the same problem too. But I using detection of P-glycoprotein expression to confirm the resistance of cell by using flow cytometer. Unfortunately the expression would not show even after 3 mounts of Tam treated cell
