I am current working on the project related to Nannochloropsis EPA for production of protein. By using aqueous solution at low temperature and pH lower than 5 it will increasing extraction efficiency. But, I wonder for how long the extraction should take? Does anyone assist or provide some hints for that? thank you. Peter
I suggest you to try stirring the dispersion for 30 to 60 minutes, followed by centrifugation, then re-disperse the precipitate for repeated extraction until two or three times.
Excessive exposure to strong acid or alkali may deteriorate the protein.

Thank you for your suggestion. Do you mean that during the extraction, acid or alkaline can be applied to? If yes, how much could be to avoid deteriorate of protein?

Please refer to the protein extraction method in the attached paper. They used alkaline extraction.
Take solid/liquid ration range 1/8 to 1/20 for extraction acid or alkalin.
Do you know the isolectric point of your extracted protein and the solubility according to pH ?
You can extract your protein at pH given the high solubility and after after centrifugation precipitate the protein in the extracted phase at the isoelectric point of your protein.
