I am going to construct a library in E. coli, and the number of variants is about 107. To make sure that every variant in the library is included, each plasmid should be transformed into E. coli and there will be more variants than the theoretical number. But how many colonies should I get?

In practice the number of clones must be greater because the process is random. Clarke and Carbon (1976) devised a formula which calculates the probability (P) of any DNA sequence being included in a library of random fragments where (a) is the insert size and (b) is the genome size.
Actual number of clones required (N) N = ln(1-P)/ln(1-a/b)
