I used to isolate alveolar macrophages by removing the organ,digested for 45 min at 37 °C with 2 mg/ml of type IV collagenase, and were passed through a 70-µm cell strainer. The total cell numbers in a mouse are 300-4,000,000.The yield of alveolar macrophages are pretty low, only about 80,000 cells per mouse.

I always get smaller numbers of cells than the papers I read, but I couldn't figure out what was wrong and how I could improve the yield.Maybe there are some differences between species of mice, I guess. I will further view more literature to improve the experimental method for observation.
