we are using EDC for activation of COOH groups and after conjugation with amine group (reaction is 12h in fridge and dark) we want to block rest of COOH. In Hermanson is used EDC and Tris/ethanolamine. Is it posible that groups are still active from first EDC or is this activation too undtable? Thank you

In my opinion, the activation lasts until there are no more nucleophilic molecules. However, if prolonged, the activation may lead to side reactions such as racemization on the alpha carbon.
On the other hand, if you just want to quench all COOH groups remaining, just add a huge amount of quencher cocktail and that´s it ;-)

It also depends on activation conditions, better to use within 10-15 minutes, if under cooling or RT to avoid complications.
