I am trying to use a 405nm laser to photo-convert different fluorescent murine tissues to track migration of immune cells. I want to avoid tissue damage and inflammation with my laser which might affect the behavior of the immune cells. Does anyone have experience with laser application and tissue inflammation?
Frist , i don't think so that 405nm,can give you any advantages, i hope you can choose other wavelength,
And about the damage effects, you can use low power laser, or even change the time or exposure, you can choose the laser dose, by different parameters, power density,spot size..

As a primer on laser induced damage to tissue, you could read the attached ICNIRP publications. Further tips: If you want to excite fluorescence, you will need to know which tissue component is excited best by what wavelength(s). There is no "one size fits all". Just choosing 405nm "because it's available" will not work or at least be sub-optimal. Do a literature search for excitation and emission wavelengths. Choose a very high optical density notch filter to block out your excitation laser.
Using pulsed excitation, and subtracting the non-excited image from the excited image will drastically improve your signal.
