I know that cycloheximide treatment prior to RT-PCR experiments is used to assess nonsense-mediated decay caused by premature stop codons. Generally if a mutation causes mRNA to be reduced at baseline, but levels are restored by CHX treatment, this is viewed as evidence for NMD... how does this mechanistically work, and are there other ways that cycloheximide treatment influences transcript levels? any thoughts/feedback greatly appreciated!
