I'm curious as to how to terminate protein expression in T7 Express Competent E. coli (high efficiency) after gene induction with 0.2% arabinose? Is there any way to halt this besides waiting for consumption of arabinose? If I have to wait for arabinose consumption, any idea on how to estimate the rate at which this occurs?
Addition of Glucose to the medium is known to repress the pBAD promoter.
I agree with Sing taht glucose can be used to repress expression. I suspect that, enough T7 Polymerase will be accumulated a few minutes after induction with arabinose and you may not able to terminate expression with addition of glucose.
To clarify, I wanted to induce protein expression of a different plasmid using IPTG. The addition of glucose would also prevent production of the second protein. Is there anyway around this?
