I want to tag a formulation made of multiple phytochemicals which constitutes flavonoids and alkaloids with 99mTc. I will be using the 99mTc tagged formulation for gamma imaging on mice or New Zealand white rabbit. Can any one tell me the detailed protocol of tagging 99mTc with phytochemicals? I read a paper at Nature Biotech which tells about tagging 99mTc to N-terminal His tags as well as purification of the tagged peptide.
It is difficult to tag 99m Tc to a flavonoids and alkaloids mixture (the formulation). Firstly, it is not possible to form chelate with these phytochemicals; secondly, covalent bond can't be established between Tc and flavonoids/alkaloids; thirdly, even if it can be tagged, these different compounds (thousands of components) will distribute to different organs of the animal, it is more than complex to decide their pharmacokinetics behaviors.
Radionuclides, 99mTc is preferred since it has high-intensity photon. It is used in Syrup of ipecacuanha Rationale Syrup of ipecacuanha that contains two alkaloids, emetine (methylcephaeline) and cephaeline. It is used to display accumulation of 99mTc-sestamibi in breast and lung tumors. 99mTc-TF has been also found to have tumor-avid properties. It is also used to detect atherothrombosis, coronary heart diseases and widely used in nuclear medicine.
