After staining intestinal organoid, what are appropriate methods (manual/automaticor computational) for quantifying the f-actin stain? I have also attached a sample PICTURE for reference. here in this picture of organoid := redcolor = Phalloidin/ f-actin and bluecolor = DAPI stain. Thank you.

Navin Ray , you can use Fiji (ImageJ) to measure actin. You will first need to split the channels so that you can work with the actin channel alone. Thresholding and then measuring the integrated density will help you get what you need.
