I am going to test the activity of AHL degrading enzyme from my Bacillus isolate. How do you measure the activity besides using the HPLC method? Is it appropriate if I apply the method from the publication below? In this publication, activity of inhibition of quorum sensing from plant extract was measured with the relative concentration of violacein produced by Chromobacterium violaceum using spectrophotometry (OD585),

Chromobacterium CV026 is suitable for detecting short chain AHLs and can not detect long chain AHLs. So you can use C6-HSL as standard AHLs and spectrophotometry for measuring AHLs degradation.
