For the isolation of particular compound from the selected extract, firstly you have select different mobile phases with varying concentration in the screening program based on the available data on your plant or extract. Then you have to select one in which separation of your compound is clear on the TLC plates.

Then you can perform HPTLC for the screening of isolated compound and further characterize it by the help of IR, NMR, and Mass spectrometric techniques.

Your question raises many subquestions. Such as 1. What plant, 2. What quantity 3. What purity 4. What costs? Plants carry many different types of molecules. As a rule of thumb a. extract the soluble one and b then separate the mixture by Chromatography. (TLC, Flash, semi prep, Prep, or production scale) If you want to have further help you have to answer the above questions.

Increasing the consumption of fruits and vegetables has been recommended by many agencies and health care systems throughout the world.

3. Are you sure that the constituent is present in it?

4. If yes, How much?

5. Whethet it is expected to be major constiuent or minor constituent in the plant matetial?

6. As a first step which solvent you have used to obtain Crude Extract?

7. What other constituents are likely to be present in the crude extract?

Steps to extract and isolate depends on answers of all these questions.

The plant I am using is Moringa oleifera Lam. I used ethanol as my extraction solvent. I am specifically trying to isolate an isothiocyanate.

Dear Joshua. You obviously have different molecules in your extractant that have to be separated. Then the question to be answered is 1. scale. (analytical, prep, production scale). And 2. what do you aim to achieve. a. to finish off with an extract less isothiocyanate or b. do you want to harvest isocyanate? Cost is always an important issue so send me answers to the above questions and I will try to send you a relevant answer. Moringa Oleifera is used in a number of traditional types of medicines. Do you want to use the result on Human/animal or are you just doing research where toxicity is of no issue?

Dear Joshua, Ensure that ethanol is almost completely evaporated from your crude extract. Take a part of extract say for eg. 1 gm of crude extract and shake it with 100 ml diethyl ether and 100 ml distilled water in an seperating funnel. Seperate both the layers. Evaporate the solvent from each fraction. Test both the fractions for presence of compound you want to isolate. Do this and let me know which fraction gives positive test for presence of the constituent. Then it can be subjected to ferther steps.
